1,6-Hexanediol is a linear aliphatic diol. It is a white waxy solid at room temperature and is easily soluble in polar solvents such as water and alcohol. Its molecular structure contains two primary hydroxyl groups, giving it excellent reactivity and hydrophilicity.

Industrial synthesis
1,6-Hexanediol is mainly synthesized through chemical methods. Common industrial routes include:
Adipic acid esterification and hydrogenation method: Using adipic acid as raw material, it is first esterified with methanol to form dimethyl adipate, and then catalytically hydrogenated to obtain the product, with a purity of more than 99%.
Biosynthesis method: Using engineered Escherichia coli to biosynthesize from renewable raw materials such as glucose.
Core application
As an important chemical intermediate, 1,6-hexanediol is widely used:
UV paint: reacts with acrylic acid to form HDDA, which is an excellent reactive diluent in UV paint.
Polyurethane: Used as a chain extender, it can significantly improve the water resistance, mechanical strength and corrosion resistance of polyurethane materials.
Polyester and plasticizer: used to synthesize high-performance polyester and can be used as a plasticizer to improve the flexibility of the material.
New role in life sciences: a key tool for phase separation research
In recent years, 1,6-hexanediol has become a central tool in the study of liquid-liquid phase separation (LLPS), which can effectively dissolve biomolecular aggregates driven by weak hydrophobic interactions. Through this property, scientists can reveal the dynamic regulation mechanism of membrane-less organelles in cells.
Release of BLM helicase and induction of homologous recombination
Studies have shown that 1,6-hexanediol dissolves "assemblyosomes" that store DNA repair proteins, releasing BLM helicase. This induces homologous recombination (HR), manifested by a significant increase in recombination events on the fly's wings. However, in the absence of guidance from DNA damage (such as X-rays), random recombination may increase cytotoxicity [1].
Affects kinase and phosphatase activities
This is a crucial reminder: 1,6-hexanediol directly and potently inhibits kinase and phosphatase activity. Significant inhibition was observed at a concentration of 1%, with almost complete inactivation of these enzymes at 5%. This inhibitory effect is independent of its function to dissolve LLPS [5].
Inhibit tumor angiogenesis
1,6-hexanediol was found to significantly inhibit pathological angiogenesis. In various models, it can reduce the formation of new blood vessels, and the mechanism may be through down-regulating the expression of cyclin A1 (Cyclin A1) at the transcriptional level [3].
Synergistic anticancer effect with Pitstop-2
Combining 1,6-hexanediol with Pitstop-2 shows powerful synergistic cytotoxicity against highly aggressive or drug-resistant non-small cell lung cancer (NSCLC) cells. This combination can also significantly inhibit the migration ability of cancer cells and has less impact on normal cells [4].
Rapidly condenses chromatin
In addition to solubilizing LLPS, 1,6-hexanediol causes a high degree of chromatin condensation and a significant reduction in motility, and can freeze chromatin even at low concentrations, an effect that is independent of its lytic activity on droplets [2].
Detection method
According to the national standard GB/T 30305-2013**, the quality detection method of industrial 1,6-hexanediol is mature:
Content determination: Gas chromatography (GC) is recommended.
Moisture determination: carried out in accordance with **GB/T 6283**.
Migration detection: liquid chromatography-tandem mass spectrometry (LC-MS/MS) can be used.
Important notes
Data interpretation requires caution: the inhibitory effect of 1,6-hexanediol on kinases/phosphatases may complicate the results of LLPS experiments [5].
Concentration is key: Dissolving LLPS usually requires high concentrations of 5%-10%, but enzyme activity is severely impaired at this concentration.
Cytotoxicity: High concentration or prolonged treatment will significantly reduce cell viability.
Non-specific effects: In addition to the above effects, it also has a significant impact on chromatin status [2]. 2,5-hexanediol, which has a similar structure, does not have this effect, indicating that its effect is specific.
Summary
1,6-hexanediol is a "star molecule" that crossed over from the industrial field and has a unique position in life sciences (especially LLPS research).
